摘要: Significant advances have been made in studies on DNA synthesis, cell cycle traverse and proliferation through the use of radiolabeled precursors. The tritiated thymidine to label cells actively synthesizing enabled Howard Pelc (1953) subdivide G1, S, G2/M phases. capability for following labeled S phase subsequent phases has also dramatically improved investigations kinetics division patterns. Recently, 5-bromodeoxyuridine (BrdUrd), base analog thymidine, substituted radioactive precursor. BrdUrd several advantages that (a) it can be more safely utilized would prohibit or limit precursors, (b), methods assaying BrdUrd, approaching sensitivity detection triatiated are currently available. One most widely used cytological technique BrdUrd-substituted is immunofluorescent assay developed by Grantzer (1982). This method requires partial denaturation single stranded conformation, heat acid treatment, provide access incorporated antibody, which usually tagged with green fluorochrome, fluorescein isothiocyanate (FITC).
