Th1 and Th2 cytokines exert regulatory effects upon islet microvascular areas in the NOD mouse

摘要: In this study, we show that intra- and peri-islet microvascular areas undergo different changes during the islet inflammation in nonobese diabetes-prone female mice. Actually, although vascular area (IVA) considerably decreases while infiltration progresses, at 15 weeks of age, bed is unexpectedly significantly increased. On contrary, intra-IVA decreased, due to vessel dilation. Later, by 20-25 a decrease both IVA occur, significant beta cell loss. Moreover, dramatic fall natural free radical scavenger values, which, turn, exert an influence upon vessels, observed. These effects are completely counteracted administration IL-4, Th2 protective cytokine; IL-10, another putative cytokine, exerts direct endothelial (EC) function, as shown increase nitric oxide synthase (eNOS) mRNA transcripts release NO vasodilatory effects; moreover, cytokine upregulates adhesion molecules on endothelia. other hand, IL-1beta, Th1 proinflammatory dramatically increases nitrite nitrate levels, well inducible (iNOS) also ICAM-1 expression circulating levels. Taken together, our findings clearly cytokines endothelia directly involved pathophysiology disease. Their reciprocal gives new insight understand role microvasculature attack.
