Differential centrifugation in culture and differentiation of rat neural stem cells.

摘要: (1) The study of neural stem cells (NSC) has attracted much attention in recent years because their therapeutic potential. However, the problem culture and differentiation NSC was how to obtain single cell suspension that preserves function NSC, remove debris caused by mechanical dissociation. In present study, we try find a simple effective way address problem, i.e. differential centrifugation. (2) After gentle dissociation using Pasteur pipette, first centrifuged at 100 g for 5 min, then recentrifuged 400 6 min. Finally, two deposits were resuspended seeded into flask respectively. from second deposit allowed further differentiation. Immunofluorescence technique used identify cell, neuron, astrocyte, oligodendrocyte. (3) centrifugation, obtained with 2-3 clusters, not only grew form neurospheres, but also differentiated neurons, astrocytes, oligodendrocytes. (4) Differential centrifugation is suspension, which will help make large-scale production neurodifferentiated more effective.
