MicroRNA Profiling Reveals Unique miRNA Signatures in IGF-1 Treated Embryonic Striatal Stem Cell Fate Decisions in Striatal Neurogenesis In Vitro

摘要: The striatum is considered to be the central processing unit of basal ganglia in locomotor activity and cognitive function brain. IGF-1 could act as a control switch for long-term proliferation survival EGF + bFGF-responsive cultured embryonic striatal stem cell (ESSC), while LIF imposes negative impact on proliferation. IGF-1-treated ESSCs also showed elevated hTERT expression with demonstration self-renewal trilineage commitment (astrocytes, oligodendrocytes, neurons). In order decipher underlying regulatory microRNA (miRNA)s IGF-1/LIF-treated ESSC-derived neurogenesis, we performed in-depth miRNA profiling at 12 days vitro analyzed candidates using Partek Genome Suite software. annotated fingerprints delineated differential expressions miR-143, miR-433, miR-503 specific treatment. Similarly, LIF-treated demonstrated miR-326, miR-181, miR-22, they were nonsignificant IGF-treated ESSCs. Our findings revealed important mRNA targets suggested interactomes. above studies introduced new genre ESSC-based neuroregenerative therapeutic applications.
