摘要: The BMP/Smad signaling pathway plays an important role in the viability and differentiation of osteoblast; however, it is not clear whether this involved fluoride-induced osteoblast differentiation. In study, we investigated osteoblast-like Saos-2 cells Cells were exposed to fluoride different concentrations (0, 0.1, 0.2, 0.4, 0.8, 1.6 mM), cell proliferation was determined using WST assays. expression marker genes such as osteocalcin (BGP) bone alkaline phosphatase (BALP) detected by qRT-PCR. We found that enhanced a dose-dependent manner 0.2 mM resulted higher genes. addition, immunofluorescence analysis showed promotion effects on associated with activation pathway. Expression phosphorylated Smad1/5(p-Smad1/5) fluoride. Plasmid vectors encoding short hairpin RNA (shRNA) targeting Smad4 gene used block pathway, which significantly reduced BGP BALP well their corresponding mRNA. mRNA levels after transfection remained low even presence present results reveal altered during period osteogenesis, activities p-Smad1/5 required for induced
