The existence of multipotent stem cells with epithelial-mesenchymal transition features in the human liver bud.

摘要: During early stage of embryonic development, the liver bud, arising from foregut endoderm, is beginning for formation future three-dimensional structure. While gene expression profiles associated with this developmental have been well explored, detailed cellular events are not as clear. Epithelial-mesenchymal transition (EMT) was thought to be essential cell migration in vertebrate embryo but seldom demonstrated human development. In study, we tried identify populations both stem and EMT features bud. Our situ studies show that phenotype occurs at initiation accompanied by up-regulation genes. A bud derived line (hLBSC) established, which expressed only genes specific mesenchymal cells hepatic cells, also octamer-binding protein 4 (OCT4) nanog. Placed appropriate media, hLBSC differentiated into hepatocytes, adipocytes, osteoblast-like neuron-like vitro. When transplanted severe combined immunodeficiency mice pre-treated carbon tetrachloride, engrafted parenchyma proliferated. These data suggests there EMT-like properties may play an important role spatial structure construction liver.
