Transplantation in Miniature Swine: IV. Chemical Characterization of MSLA and Ia-like Antigens

摘要: Immunochemical analyses of radioactively labeled lymphocyte antigens from miniature swine three different homozygous major histocompatibility (MHC) types, AA, CC, and DD, have been performed. Anti-MHC sera were incubated with lentil lectin purified Nonidet P-40 extracts. Antigen-antibody complexes then precipitated protein A bearing staphylococci, eluted, electrophoresed on sodium dodecyl sulfate polyacrylamide gels. Analysis AA or DD cells revealed peaks 42,000, 31,000, 25,000, 11,000 dalton m.w. Platelet absorption the anti-MHC yielded antibodies that only intermediate molecules lysed a subpopulation peripheral blood lymphocytes, suggesting these analogues murine Ia antigens. Antibodies eluted platelets all populations 42,000 peaks, indicating represent analogue H-2 antigens, containing heavy chain putative β 2 -microglobulin.
