Structure activity characterization of Bordetella petrii lipid A, from environment to human isolates.

摘要: Bordetella petrii, a facultative anaerobic species, is the only known member of genus with environmental origin. However it was also recently isolated from humans. The structures B. petrii lipid A moieties endotoxins were characterized here for first time an strain and compared to that human isolates. Characterization achieved using chemical analyses, gas chromatography-mass spectrometry, Matrix Assisted Laser Desorption Ionisation mass spectrometry. analyses revealed different contain common bisphosphorylated β-(1→6)-linked d-glucosamine disaccharide hydroxytetradecanoic acid in amide as well at C-3' ester linkages. Similar pertussis bronchiseptica lipids A, C-2' position substituted by tetradecanoic acid. Unlike B. pertussis, C-2 either 12:0 or 14:0 and/or their 2-OH forms. Depending on origin differed length degree fatty acylation impacted IL-6 TNF-α inflammatory responses tested. In one isolate we showed presence C-3 short-chain 10:0(3-OH), which according our previous more characteristic pathogens like B. pertussis parapertussis.
