Quantification of Bovine Sperm Separation by a Swim-up Method Relationship to Sperm Motility, Integrity of Acrosomes, Sperm Migration in Polyacrylamide Gel and Fertility

摘要: The number of bovine spermatozoa separated in a swim-up procedure was quantified using an electronic cell counter. In initial test the procedure, non-frozen sperm samples with different ratios live to dead cells were prepared and tested for counted by procedure. ejaculates from six bulls, swimming up related present (R2 = 0.97). Next, quality frozen-thawed semen immediately after thawing measured at 37 C count, motility, intact acrosome migration polyacrylamide gel then compared fertility used artificial insemination. Twenty-nine 11 bulls evaluated. Correlations highest on ejaculate basis motility (r 0.41, P 0.05) count 0.35, 0.06). On bull basis, had correlation 0.59, multiple regression model predict male that included all described measures quality, R2 value 0.69 obtained. This is first report showing ability swim out more dense medium (whole milk-glycerol extender) into culture media quantitatively vivo fertility.
