摘要: The influence of central injection a new corticotropin releasing factor (CRF) antagonist, astressin, [cyclo(30-33)[D-Phe12,Nle21,38,Glu30,Lys33]r/ hCRF12-41)], on exogenous and endogenous CRF-induced gastric ileus stimulation bowel discharges was investigated in conscious rats. Intracisternal (ic) CRF (0.6 microg) reduced emptying noncaloric solution to 17.1 +/- 4.9% compared with 50.1 4.6% control group injected i.c. vehicle. Astressin (1,3 10 microg, i.c.) dose dependently prevented ic delayed by 33, 100 100%, respectively, had no effect basal emptying. Abdominal surgery cecal manipulation (1 min) 19.8 5.5% 3 hr postsurgery 59.9 5.2% after anesthesia alone plus postoperative 56, 93 92%, respectively. Intracerebroventricular water-avoidance stress stimulated pellet output (number/60 5 1 11 2, fecal i.c.v. vehicle exposure stress. (3 i.c.v.) blocked action 47 63%, colonic response 0 54%, These data indicate that astressin into the CSF at low doses (1-10 has an antagonistic against stress-related alterations gastrointestinal motor function, without intrinsic these vivo systems. may be useful tool explore functional CRF-dependent physiological pathways specific brain nuclei.
