摘要: Phosphoserine has potential effectiveness as a simple substrate in preparing bone replacement materials, which could enhance forming ability. However, there is need to investigate the independent effect of phosphoserine on osteogenic differentiation human adipose stem cells (hADSCs). hADSCs were cultured an medium with phosphoserine. Cell proliferation was analysed by CCK8 and measured alkaline phosphatase (ALP) activity, von Kossa staining real time-polymerase chain reaction (RT-PCR). No stimulatory cell noted at Days 1, 4 7. Deposition calcium increased after addition mRNA expression type I collagen (COL-I), (ALP), osteocalcin (OCN), Osterix, morphogenetic protein-2 (BMP-2) RUNX2 markedly treatment. The BMP-2 antagonist, noggin, its receptor kinase inhibitors, dorsomorphin LDN-193189, attenuated phosphoserine-promoted ALP activity. BMP-responsive Runx2-responsive reporters activated Thus can promote hADSCs, probably activating BMP Runx2 pathways, be promising approach for enhancing capacity cell-based construction tissue engineering.
