Role of endothelium and nitric oxide in modulating in vitro responses of colonic arterial and venous rings to vasodilatory neuropeptides in horses

摘要: Abstract The objective of this study was to determine and compare the in vitro responses equine large colon arterial venous rings vasodilatory neuropeptides; calcitonin gene-related peptide (CGRP); substance P (SP); vasoactive intestinal polypeptide (VIP); acetylcholine (ACh), a standard nonpeptide endothelium-dependent vasodilator. Responses vessel graded con-centrations (10 11 M 10 5 M) each drug were determined endothelium-intact, denuded, N  -nitro-L-arginine methyl ester (L-NAME, M)-treated that pre-contracted with norepinephrine. Percentage maximal relaxation (PMR), defined as % decrease from contracted state, determined. Because all did not relax at least 50%, EC 50 values could be consistently calculated. Arterial intact endothelium more sensitive CGRP, compared VIP SP, conditions than CGRP or SP. Overall, arteries had greater PMR for ACh SP VIP. Intact L-NAME treated denuded arteries; there no differences arteries. Veins ACh. Calcitonin caused arteries, whereas causes veins. relax-ation dependent upon presence endothelium. response veins among tested different, suggesting has direct actions on smooth muscle.
