摘要: The steroid hormone ecdysone, which controls insect molting and metamorphosis, is synthesized in the prothoracic gland (PG), several steroidogenic enzymes that are expressed specifically PG involved ecdysteroidogenesis. In this study, we identified new regulators transcriptional control of silkworm enzyme genes. silico analysis predicted potential cis-regulatory elements (CREs) for homeodomain transcription factors Antennapedia (Antp) POU-M2 proximal promoters Antp dynamically during larval development, their overexpression embryo-derived (BmE) cells induced expression Importantly, luciferase reporter analyses, electrophoretic mobility shift assays, chromatin immunoprecipitation assays revealed promote gene Phantom (Phm) by binding directly to specific motifs within overlapping CREs Phm promoter. Mutations these promoter suppressed activities both BmE decreased mutated PG. addition, pulldown co-immunoprecipitation demonstrated can interact with POU-M2. Moreover, RNA interference-mediated down-regulation either or wandering not only ecdysone titer but also led failure metamorphosis. summary, our results suggest coordinate directly, indicating roles proteins regulating
