摘要: Abstract The Nile tilapia Sox9b gene is characterized as a homolog of the mammalian Sox9 gene, which exhibits testis-biased expression and involved in testis development. However, transcriptional regulation poorly understood. In this study, we demonstrated that male sex differentiation doublesex mab-3 related transcription factor 1 (Dmrt1) was predominantly expressed Dmrt1 knockdown decreased testis. An silico analysis predicted proximal promoter had two potential cis-regulatory elements (CREs) for factor. Together, luciferase reporter site-directed mutagenesis revealed increased activity via specific CRE near translation start site. A chromatin immunoprecipitation (ChIP) an electrophoretic mobility shift assay (EMSA) confirmed could directly bind to promoter. Taken together, our results demonstrate sex-differentiation positively regulates by binding within
