摘要: Long-read sequencing technologies were launched a few years ago, and in contrast with short-read technologies, they offered promise of solving assembly problems for large complex genomes. Moreover by providing long-range information, it could also solve haplotype phasing. However, existing long-read still have several limitations that complicate their use most research laboratories, as well and/or genome projects. In 2014, Oxford Nanopore released the MinION® device, small low-cost single-molecule nanopore sequencer, which offers possibility long DNA fragments. The reads generated using instrument is challenging assemblers not implemented to deal exhibiting close 30% errors. Here, we presented hybrid approach developed take advantage data device. We sequenced well-known bacterium, Acinetobacter baylyi ADP1 applied our method obtain highly contiguous (one single contig) accurate even repetitive regions, an Illumina-only assembly. Our strategy was able generate NaS (Nanopore Synthetic-long) up 60 kb aligned entirely no error reference spanned conserved regions. average accuracy reached 99.99% without losing initial size input reads. described tool, allowing microbial genomes method, based ideally on 20x 50x Illumina respectively, provides efficient cost-effective way or eukaryotic very short time facilities. Moreover, demonstrated although technology relatively new technology, currently high rate, already useful generation high-quality assemblies.
