Extended safety and efficacy studies of a live attenuated double leucine and pantothenate auxotroph of Mycobacterium tuberculosis as a vaccine candidate.

摘要: We have previously described the development of a live, fully attenuated Mycobacterium tuberculosis (Mtb) vaccine candidate strain with two independent attenuating auxotrophic mutations in leucine and pantothenate biosynthesis. In present work, those studies been extended to include testing for protective efficacy long-term guinea pig survival model safety highly susceptible Rhesus macaque. To ΔleuD ΔpanCD HIV-infected human populations, Simian immunodeficiency virus (SIV)-infected macaque group was included. Immunization non-replicating conferred protection against challenge virulent M. equivalent that afforded by BCG as measured survival. studies, clinical, hematological bacteriological monitoring both SIV-positive SIV-negative macaques immunized ΔpanCD, revealed no vaccine-associated adverse effects. The results support further viable (TB) candidate.
