Tetrodotoxin-sensitive Na+ channels and muscarinic and purinergic receptors identified in human erythroid progenitor cells and red blood cell ghosts

摘要: This article concerns the identification of different types voltage-gated Na+ channels and muscarinic purinergic receptors that are expressed in human erythroid precursor cells red cell ghosts. We analyzed, by RT-PCR, RNA was extracted from purified synchronously growing progenitor cells, differentiating erythroblasts to reticulocytes 7 14 days. These extracts were free white platelet contamination. Two voltage-gated, tetrodotoxin-sensitive found. Nav1.4 Nav1.7, former known be present skeletal muscle latter peripheral nerve. By using a pan channel antibody Western blotting, an immunoreactive detected ghosts blood consistent with expression these two channels. The transcripts for four five subtypes also identified, including M2, M3, M4, M5, whereas subtype M1 not Expression type P2X1, P2X4, P2X7, P2Y1 P2Y2, P2Y4, P2Y6 searched but did find hBNP-1, 1b brain sodium phosphate cotransporter, cystic fibrosis transmembrane conductance regulator (CFTR). Implications regarding presence their functional significance discussed.
