摘要: Background The efficiency of RNA interference (RNAi) varies considerably among different insect species, and there is growing evidence to suggest that degradation double-stranded (dsRNA) prior uptake an important factor limits the RNAi in insects. In Locusta migratoria, highly efficient when dsRNA delivered by injection, but not feeding. However, detailed mechanisms causing such differential are still elusive. Results We identified characterized full-length complementary DNAs (cDNAs) two new nuclease (dsRNase) genes from L. which were named LmdsRNase1 LmdsRNase4. Transcript analyses revealed LmdsRNase4 expressed hemolymph with relatively lower expression other tested tissues. Our study using heterologously fusion proteins showed can degrade rapidly at optimal pH 5, whereas had no activity any values examined. comparing substrate specificity four LmdsRNases, we found only LmdsRNase2 digested dsRNA; however, our experiments suggested physiological (7.0) suppresses permitting significant stability this tissue. Conversely, midgut juice (6.8) ideal for activity, resulting midgut. Conclusion tissues or compartments significantly alter influencing LmdsRNase migratoria.
