摘要: Molecular tools of the intracellular protozoan pathogens Apicomplexa and Kinetoplastida for manipulation host cell machinery have been focus investigation approximately two decades. Microsporidia, fungi-related microorganisms forming another large group obligate parasites, are characterized by development in direct contact with cytoplasm (the majority species), strong minimization machinery, acquisition unique transporters to exploit metabolic system. All aforementioned features suggestive ability microsporidia modify regulatory pathways. Seven proteins microsporidium Antonospora (Paranosema) locustae predicted signal peptides but without transmembrane domains were overexpressed Escherichia coli. Western-blot analysis antibodies against recombinant products showed secretion parasite from different functional categories into infected cell. Secretion hexokinase α/β-hydrolase was confirmed immunofluorescence microscopy. In addition, this method specific accumulation A. nuclei. Expression hexokinase, trehalase, leucine-rich repeat any exogenous peptide led their yeast Pichia pastoris. contrast, not found culture medium, though a significant amount enzyme accumulated membrane fraction. These results suggest that possess broad set enzymes secreted cells control processes molecular programs.
