Comparison of Picrosirius Red Staining With Second Harmonic Generation Imaging for the Quantification of Clinically Relevant Collagen Fiber Features in Histopathology Samples

摘要: Stromal collagen alignment has been shown to have clinical significance in a variety of cancers and other diseases accompanied by fibrosis. While much the biological importance changes demonstrated using second harmonic generation (SHG) imaging experimental settings, implementation into routine pathology practice is currently prohibitive. To translate assessment organization workflow, surrogate visualization method needs be examined. The objective present study was quantitatively compare metrics generated from SHG microscopy commonly available picrosirius red stain with standard polarization (PSR-POL). Each technique compared established image segmentation fiber tracking algorithms human pancreatic cancer as model, which characterized pronounced stroma reorganized fibers. Importantly, PSR-POL produced similar quantitative trends for most benign cancerous tissues measured SHG. We found it notable that detects higher counts, alignment, length, straightness, width but still correlates well results. may provide sufficient additional information conventional laboratory certain types quantification.
