Organochlorine pesticides induce epithelial to mesenchymal transition of human primary cultured hepatocytes

摘要: Persistent organic pollutants (POPs) are a group of or chemicals that adversely affect human health and persistent in the environment. These highly toxic compounds include industrial chemicals, pesticides such as organochlorines, unwanted wastes dioxins. Although studies have described general toxicity effects organochlorine pesticides, mechanisms underlying its potential carcinogenic liver not well understood. In this study, we analyzed effect three (dichlorodiphenyltrichloroethane, heptachlore endosulfan) 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) on epithelial to mesenchymal transition (EMT) primary cultured hepatocytes. We found these modified hepatocyte phenotype, inducing cell spread, formation lamellipodia structures reorganization actin cytoskeleton stress fibers. morphological alterations were accompanied by disruption cell-cell junctions, E-cadherin repression albumin down-regulation. Interestingly, characteristic features dedifferentiating hepatocytes correlated with gain expression various genes, including vimentin, fibronectin receptor ITGA5. results show organochlorines TCDD accelerate dedifferentiation EMT processes. events could account, at least part, for carcionogenic and/or fibrogenic activities POPs.
