Engulfment of Apoptotic Cells Is Negatively Regulated by Rho-mediated Signaling

摘要: The rapid and efficient phagocytosis of apoptotic cells plays a critical role in preventing secondary necrosis, inflammation as well tissue remodeling regulating immune responses. However, the molecular details engulfment are just beginning to be elucidated. Among Rho family GTPases, previous studies have implicated for Rac Cdc42 uptake by phagocytes, yet has remained unclear. Here, we present evidence that Rho-GTP levels decrease during engulfment. RhoA seems negatively affect basal engulfment, such inhibition Rho-mediated signaling phagocytes enhanced targets. Activation endogenous or overexpression constitutively active forms also inhibited By testing mutants selectively activate downstream effectors, Rho-kinase seemed primarily responsible this inhibitory effect. Taken together, these data suggest Rho- Rho-kinase-mediated might important which could implications several clinical trials involving kinase.
