摘要: Helicases and translocases use the energy of nucleoside triphosphate binding hydrolysis to unwind/resolve structured nucleic acids or move along a single-stranded double-stranded polynucleotide chain, respectively. These molecular motors facilitate variety transactions including replication, DNA repair, recombination, transcription. A key partner eukaryotic helicases/translocases is protein Replication Protein (RPA). Biochemical, genetic, cell biological assays have demonstrated that RPA interacts with these human physically functionally, their association enriched in cells undergoing replication stress. The roles are orchestrated pathways acid metabolism. stimulates helicase-catalyzed unwinding, enlists sites action, modulates activities fork remodeling, checkpoint activation, telomere maintenance. dynamic interplay between just beginning be understood at cellular levels, there still much learned, which may inform potential therapeutic strategies.
