摘要: 1. The objective of the present study was to examine role endothelial selectins (i.e. P- and E-selectin) in leukocyte-endothelium interactions colonic venules by use intravital microscopy. 2. Balb/c mice were exposed dextran sodium sulphate (DSS) drinking water for 5 days or treated intraperitoneally (i.p.) with tumour necrosis factor-alpha (TNF-alpha) 3 h. 3. In DSS-treated mice, mRNA both E-selectin expressed leukocyte rolling adhesion increased 27+/-3 cells min(-1) 36+/-8 mm(-1), respectively. An anti-P-selectin antibody abolished DSS-induced rolling, whereas an against had no effect. Established insensitive inhibition selectins. 4. DSS markedly production TNF-alpha colon. 22+/-3 45+/-4 mm(-1). Only P-selectin significantly reduced (>94%) provoked TNF-alpha. Leukocyte not changed late treatment. contrast, pretreatment only but also completely inhibited firm response 5. This demonstrates that plays important venules, experimental colitis when stimulated Moreover, P-selectin-dependent found be a precondition TNF-alpha-induced adhesion. Thus, these findings suggest may key target reduce pathological recruitment inflammatory
