LSD1 Interacts with Zfp516 to Promote UCP1 Transcription and Brown Fat Program

摘要: Zfp516, a brown fat (BAT)-enriched and cold-inducible transcription factor, promotes of UCP1 other BAT-enriched genes for non-shivering thermogenesis. Here, we identify lysine-specific demethylase 1 (LSD1) as direct binding partner Zfp516. We show that, through interaction with LSD1 is recruited to BAT-enriched genes, such PGC1α, function coactivator by demethylating H3K9. also that induced during adipogenesis its activity required the BAT program. Furthermore, ablation in mice using Myf5-Cre alters embryonic development. Moreover, BAT-specific deletion via use UCP1-Cre impairs program development, making resemble WAT, reducing thermogenic promoting obesity. Finally, demonstrate an in vivo requirement Zfp516-LSD1 gene activation.
