Characterization of a novel (R)-mandelate dehydrogenase from Pseudomonas putida NUST506

摘要: Abstract ( R )-Mandelate dehydrogenase (RMDH) has the potential to produce chiral mandelic acid. The present work reports isolation and identification of a Pseudomonas putida NUST506 contained RMDH. strain was rod shaped with polar flagella, approximately 0.6–0.9 μm wide 1.7–2.3 μm long. RMDH purified from strain. molecular weight enzyme calculated be 61 kDa. optimal conditions for were pH 8.5 30 °C. At optimum condition, K m k cat 2.0 × 10 −2  mM 0.9 s −1 )-mandelic acid, 1.8 × 10 NAD + , as well 1.5 × 10 0.3 s NADP respectively. activity increased by dithiothreitol (DTT), but obviously inhibited Zn 2+ Hg sodium dodecyl sulfate (SDS) ethylenediamine tetra-acetic acid (EDTA).
