Intersectional Gene Expression in Zebrafish Using the Split KalTA4 System

摘要: In this study, we describe the adaptation of split Gal4 system for zebrafish. The Gal4-UAS is widely used expression genes-of-interest by crossing driver lines expressing transcription factor (under control promoter interest) with reporter where upstream activating sequence (UAS) repeats (recognized Gal4) drive genes-of-interest. Split system, hemi-drivers separately encode DNA-binding domain (DBD) and activation (AD) Gal4. When encoded under two different promoters, only those cells in intersection promoters' pattern which both promoters are active reconstitute a functional activate from UAS-driven transgene. We zebrafish-optimized version Gal4, KalTA4, generated hemi-driver encoding KalTA4 DBD KalTA4's AD. show that domains can assemble vivo transactivate UAS transgene each alone cannot reporter. Also, transactivation happen several cell types, similar efficiency to intact KalTA4. Finally, transient mosaic assays, when preceded distinct they restrict an broader (sox10) its constituent smaller neuronal pattern. should be useful intersectional manner, allowing more refined manipulations populations
