Nerve growth factor induces survival and differentiation through two distinct signaling cascades in PC12 cells

摘要: Nerve growth factor induces differentiation and survival of rat PC12 pheochromocytoma cells. The activation the erk cascade has been implicated in transducing multitude signals induced by NGF. In order to explore role this signaling NGF mediated survival, proliferation, we generated recombinant adenoviruses which express intermediates their wild type, dominant negative constitutively activated forms. We show that cells requires activity ras/erk pathway, whereas inhibition pathway had no effect on or proliferation. Constitutively active forms ras, raf mek cell differentiation, while interfering inhibited differentiation. Survival serum-free medium did not require pathway. Instead, PI3 Kinase was necessary for survival. Interestingly, versions were able promote but again dependent Kinase. Therefore, at least two distinct pathways are required mediation functions.
