Andrographolide protects against lipopolysaccharide-induced vascular hyporeactivity by suppressing the expression of inducible nitric oxide in periaortic adipose

摘要: This study investigated the role of perivascular adipose tissue (PVAT) in beneficial effects andrographolide on vascular reactivity endotoxaemic rats. After being challenged by lipopolysaccharide (4 mg/kg intraperitoneally), rats were treated with (1 intraperitoneally). The response to phenylephrine aortic rings or without PVAT was recorded. Vascular relaxing effect determined bioassay experiments. Inducible nitric oxide synthase (iNOS) tested Western blot, immunofluorescence, and quantitative polymerase chain reaction. Lipopolysaccharide injection lowered contraction force induced treatment reversed these effects. In experiments, transferring bathing solution incubated a PVAT+ ring PVAT- relaxation recipient. from more potent than vehicles. Andrographolide decreased levels iNOS protein messenger RNA significantly higher Our results suggest that restores downregulation PVAT.
