摘要: Despite increasing evidence to suggest that microRNA (miR)-146a and miR-181b are involved in the regulation of immune responses tumor progression, their roles silicosis remain be fully elucidated. Therefore, present study examined miR-146a inflammatory responses, effect on expression necrosis factor-α (TNF-α) interleukin-1β (IL-1β) chemokines silicon dioxide (SiO2)-induced NR8383 rat macrophages. Alterations levels rats with have been previously investigated using miRNA arrays. In study, were assessed reverse transcription-quantitative polymerase chain reaction (RT-qPCR). The cells transfected miRNA-146a mimics or inhibitors, culture supernatants collected following SiO2 treatment for 12 h. TNF-α IL-1β detected western blotting, RT-qPCR ELISA. Analysis variance Student's two-tailed t-test used perform statistical analyses. level was significantly increased, while decreased fibrotic lungs silicosis, compared normal rats. It observed that, h, increased knockdown knockdown, inhibitor-treated controls (P<0.05). By contrast, mimic transfection led a significant reduction (P<0.05), responsible decrease results provide supporting pathogenesis they may candidate therapeutic target this disease.
