摘要: The aim of this study was to investigate the antitumor activities Phyllanthus amarus (PHA) and its potential herb–drug interactions with 5-Fluorouracil (5-FU). Cell viability, ribonucleotides (RNs) deoxyribonucleotides (dRNs) levels, cell cycle distribution, expression thymidylate synthase (TS) ribonucleotide reductase (RR) proteins were measured 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, high performance liquid chromatography tandem mass spectrometry (HPLC/MS/MS) method, flow cytometry Western blot analysis, respectively. Our standardized PHA extract showed toxicity HepG2 cells at concentrations after 72 h exposure induced G2/M arrest. Combined use 5-FU resulted in significant decreases ATP, CTP, GTP, UTP dTTP while AMP, CMP, GMP dUMP levels increased significantly compared alone. Further, could increase role arrest S phase by 5-FU. Although alone had no direct impact on TS RR, change RNs dRNs when combined This may be due or regulation key enzyme steps intracellular metabolism.
