摘要: The rapid interconversion of carbon dioxide and bicarbonate (hydrogen carbonate) is catalysed by metalloenzymes termed carbonic anhydrases (CAs). CAs have been identified in all three domains life can be divided into five evolutionarily unrelated classes (α, β, γ, δ and ζ) that do not share significant sequence similarities. function the mammalian, prokaryotic plant α-CAs has intensively studied but filamentous ascomycetes mostly unknown. ascomycete Sordaria macrospora codes for four CAs, β-class one α-class. Here, we present a functional analysis CAS4, S. α-class CA. CAS4 protein was post-translationally glycosylated secreted. knockout strain Δcas4 had significantly reduced rate ascospore germination. To determine cas genes required S. macrospora growth under ambient air conditions, constructed double triple mutations possible combinations quadruple mutant. Vegetative mutant lacking drastically compared to wild type invaded agar normal conditions. Likewise fruiting bodies were embedded completely devoid mature ascospores.
