摘要: We report the application of NMR dynamic spectral editing for probing structure and dynamics molecular constituents in fresh, intact cells freshly prepared thylakoid membranes Chlamydomonas reinhardtii (Cr.) green algae. For isotope labeling, wild-type Cr. were grown on 13C acetate-enriched minimal medium. 1D J-coupling based dipolar-based MAS spectra applied to distinguish resonances different components. recorded over a physiological temperature range, whole-cell compared those taken from membranes, evaluating their composition dynamics. A theoretical model polarization transfer was used simulate relative intensities direct, J-coupling, which degree lipid segmental order rotational acyl chains estimated. observe that signals dominate profile whole algae cells, demonstrating with our novel method, membrane characteristics can be detected atomistic precision inside photosynthetic cells. The experimental procedure is rapid applicable fresh cell cultures, could as an original approach detecting chemical profiles, vivo functional states.
