摘要: N-Vinylpyrrolidone-2 (NVP) is a monomeric compound used as an industrial intermediate. Nine of 11 studies previously reported involved exposure rats (two different strains), mice or hamsters to NVP by the inhalation route at concentrations up 120 ppm (6 hr/day, 5 days/wk) over period 1 wk 12 months. The remaining two through drinking water gavage dose levels 100 mg/kg body weight/day. Reduced weight gain was seen in exposed more for 3 months and 45 only day. Effects were on haematological (reduced haemoglobin, erythrocyte count, haematocrit) clinical chemistry parameters (specially raised γ-glutamyltransferase activity decreases plasma protein), liver increase lesions (centrilobular single-cell necrosis foci hepatocellular alteration) but not hamsters. Rats 40 weight/day developed similar changes. Atrophy olfactory epithelium hyperplasia nasal respiratory 7 response 13 (no observed-adverse-effect level, NOAEL). These indicated that upper tract are main targets toxicity.
