P2X7R-dependent regulation of glycogen synthase kinase 3β and claudin-18 in alveolar epithelial type I cells of mice lung

摘要: The purinergic receptor P2X7 represents an ATP-gated ionotropic with a selective localization in alveolar epithelial type I cells of the lung. Despite involvement inflammatory processes lung, it is not established whether this plays specific role cell biology. There evidence that influences Wnt/β-catenin signalling pathways under conditions injury. Here, we investigated expression GSK-3β, potent protein kinase involved barrier functions, and tight junction molecules occludin, claudin-4 claudin-18 wild-type P2X7−/− mice. Western blot analysis, immunohistochemistry quantitative real-time RT-PCR revealed remarkable increase mRNA lungs mice animals. Furthermore, from animals showed decreased levels GSK-3β its inactive form (pS9). Conversely, knockout exhibited transcript abundance as measured by microarray PCR. Our data are consistent hypothesis P2X7R contributes to function through effects on GSK-3β. these suggest potential reciprocal regulation epithelium.
