Human schistosomiasis haematobium: effective diagnosis of active infection using a pair of monoclonal antibodies against soluble egg antigen.

摘要: The present study was designed to prepare monoclonal antibodies (MAbs) against Schistosoma haematobium soluble egg antigen (SEA) with immunodiagnostic potential for urinary schistosomiasis. From a panel of MAbs, pair IgG1 MAbs (2D/11C and 10B/2C) specific S. SEA selected. Both recognized one band 42-kDa molecular weight by western blots. employed in sandwich ELISA the detection circulating schistosome (CSA), as antigen-capturing antibody other peroxidase-conjugated antigen-detecting antibody. lower limit assay 1 ng/ml SEA. performed on sera 65 haematobium-infected patients, 25 patients infected parasites (Fasciola hepatica, Echinococcus granulosus), 20 noninfected individuals. CSA demonstrated 89% group. However, negative healthy individuals parasites, giving an overall specificity 100% assay. A positive correlation (r=0.37, p<0.01) detected between number eggs excreted 10 ml urine level patients. Our data show that use anti-S. provides sensitive method immunodiagnosis active Moreover, using this MAb/ELISA system proven correlate intensity infection hence morbidity assessment.
