HIV-1 infection and pathogenesis in a novel humanized mouse model

摘要: The Rag2-γC double-knockout (DKO) mouse lacks T, B, and natural killer (NK) cells, allows development of a functional human immune system with CD34+ hematopoietic stem/progenitor cells (DKO-hu HSCs). Normal dendritic are present in peripheral blood, thymus, spleen, lymph nodes. We report that both CCR5 CXCR4 expressed on immature mature T cells. DKO-hu HSC mice allow efficient HIV-1 infection plasma high viremia. High levels productive occur the Human CD4+ gradually depleted by dose-dependent manner. In addition, persists infected for at least 19 weeks, infectious lymphoid tissues. Thus, can serve as relevant vivo model to investigate mechanisms immunopathogenesis well develop anti–HIV-1 therapeutics.
