摘要: Over the last two decades, several humanized mouse models have been used to experimentally analyze function and development of human immune system. Recent advances lead establishment new murine-human chimeric with improved characteristics, both in terms engraftment efficiency situ multilineage hematopoietic development. We describe here use newborn BALB/c Rag2(-/-)gamma(c) (-/-) mice as recipients progenitor cells produce "human system" (HIS) (BALB-Rag/gamma) mice, using fetal liver progenitors. The major subsets dendritic cell lineage, namely, BDCA2(+)CD11c(-) plasmacytoid BDCA2(-)CD11c(+) conventional cells, can be found HIS mice. In order manipulate expression genes interest, genetically engineered ex vivo by lentiviral transduction before performing xenograft transplantation. Using this model, system assessed for fundamental pre-clinical purposes.
