AngleJ: A new tool for the automated measurement of neurite growth orientation in tissue sections

摘要: Abstract Background Regeneration of axons is one means to restore function after central nervous system and peripheral injury. Besides increasing the number regenerating axons, guidance over long distances into across a lesion site are important determinants for efficient functional restoration. Quantification axon growth directions therefore an measure efficacy neuroregenerative approaches. While several methods exist manually or automatically trace neurites in images neuronal cultures determine their length, tools effect neurite tissue sections do not exist. New method Because manual measurements orientation labor-intensive, time-consuming unreliable, plugin called AngleJ open source software ImageJ was developed that determines axonal immunohistochemically labeled spinal cord. Results Given user-defined filters thresholds, accurately detects intact cord white matter hemisection model measures distribution pattern angles. Comparison with existing Values measured angles strongly correlate obtained by (Pearson correlation 0.88–0.97 0.76–0.94 sprouting site). Conclusions can be used as fast alternative angle measurement conjunction its code freely available community.
