摘要: The electrophoretic migration in polyacrylamide gels of oligogalacturonic acids (OGAs) derivatized by a fluorophore (2-aminoacridone) was studied. We found conditions such that OGAs can be separated up to degree polymerization (DP) 40. dependent on methylation and DP, because the OGA mobility relies charge galacturonic acid residues. Since both methylated unmethylated oligosaccharides resolved, polysaccharide analysis using carbohydrate gel electrophoresis (PACE) is powerful method for studying fingerprint pectin hydrolysis. It used characterize endopolygalacturonase (Endo-PG) tolerance methylation. Furthermore, an Endo-PG distinguish low highly pectin, PACE investigate blockwise or nonblockwise distribution polygalacturonic acid. show applied crude cell wall preparations Arabidopsis inflorescence stems.
