Tyrosine 766 in the fibroblast growth factor receptor-1 is required for FGF-stimulation of phospholipase C, phospholipase D, phospholipase A(2), phosphoinositide 3-kinase and cytoskeletal reorganisation in porcine aortic endothelial cells.

摘要: Fibroblast growth factor-mediated signalling was studied in porcine aortic endothelial cells expressing either wild-type fibroblast factor receptor-1 or a mutant receptor (Y766F) unable to bind phospholipase C-(γ). Stimulation of the resulted activation phospholipases C, D and A(2) increased phosphoinositide 3-kinase activity. also stress fibre formation cellular shape change. Cells Y766F failed stimulate as well 3-kinase. Furthermore, no change observed. Both activated MAP kinase elicited proliferative responses cells. Thus, mediated dependent on tyrosine 766. whilst 766 not required for response, it cytoskeletal reorganisation.
