摘要: Leao's spreading depression of cortical activity is a propagating silencing neuronal resulting from depolarization (SD). We evaluated the contributions action potential (AP) failure and adenosine A1 receptor (A1R) activation to evoked spontaneous electrocorticographic (ECoG) after SD in vivo, in anesthetized mice. compared with SD-induced effects on AP-dependent transmission, synaptic potentials transcallosal thalamocortical pathways. After SD, APs recovered rapidly, within 1-2 min, as demonstrated by distant projection targets. Evoked corticocortical postsynaptic next, ∼5 min. Spontaneous ECoG together, ∼10-15 min. The duration was shortened 20% systemic (10 mg/kg) or focal (30 µM) administration A1R competitive antagonist 8-cyclopentyl-1,3-dipropylxanthine (DPCPX). also application exogenous deaminase (ADA; 100 U/mL), conversely, prolonged 50% non-competitive ADA inhibitor deoxycoformycin (DCF; 100 µM). concluded that while initial block brief, activation, part, contributes persistent secondary phase depression.
