HLA DQA1 and Polymarker Validations for Forensic Casework: Standard Specimens, Reproducibility, and Mixed Specimens

摘要: This study describes the testing performed by Minnesota Forensic Science Laboratory (MFSL) to validate Amplitype® DQA1 and Polymarker (PM) PCR Amplification Typing Kits before implementation for casework. All studies were based on analysis of mock forensic case samples, which assembled from various biological samples individuals at MFSL. To address validation standard specimens, DNA was isolated semen, vaginal secretions, saliva, urine, blood samples. results all tissues a particular individual yielded same typing using both PM systems. Reproducibility between laboratories evaluated having duplicate analyzed second laboratory. The Roche Biomedical Laboratories (RBL) sent set cases analyses including extraction, quantitation, amplification, RBL their established procedures. laboratories, approximate 30 single source analyzed, in agreement. Mixed specimens examining obtained semen/vaginal, semen/saliva, semen/blood, semen/urine, semen/vaginal/blood mixtures. these mixtures It determined that incorporating wash step sperm cell pellet, complete separation nonsperm fraction more likely be attained.
