Sequence structure and expression of a cloned β-glucosidase gene from an extreme thermophile

摘要: The gene for a β-glucosidase from the extremely thermophilic bacterium Caldocellum saccharolyticum has been isolated genomic library and sequenced. An open reading frame identified by computer analysis of sequence could encode protein Mr 54400, which is close to size polypeptide experimentally determined using maxicells. Analysis amino-terminal residues produced in Escherichia coli suggests that it processed methionine aminopeptidase. A within C. DNA upstream was found act as promoter expression thermophile E. coli. overproduced Bacillus subtilis where retains its enzymatic activity heat stability. There appears be single copy DNA.
