CSE1L modulates Ras-induced cancer cell invasion: correlation of K-Ras mutation and CSE1L expression in colorectal cancer progression

摘要: BACKGROUND: Ras plays an important role in colorectal cancer progression. CSE1L (chromosome segregation 1-like) gene maps to 20q13, a chromosomal region that correlates with development. We investigated the association of METHODS: The effect on metastasis-stimulating activity was studied animal model tumor cells expressing CSE1L-specific shRNA and v-H-Ras. expression evaluated by immunohistochemical analysis 127 surgically resected tumors. K-Ras mutations were analyzed direct sequencing. RESULTS: knockdown reduced Ras-induced metastasis B16F10 melanoma C57BL/6 mice. v-H-Ras altered cellular trafficking increased secretion. Most tumors positive for staining (98.4%, 125 127). Colorectal mutation or high cytoplasmic correlated T status (depth penetration; P 5 .004), stage (P lymph node .019).
