摘要: Intraocular grafts of rat hippocampal tissue, grafted either directly from the immature donor brains (fresh) or after storage in liquid nitrogen at -196 degrees C (freeze-stored), were compared with regard to survivability and histological connective organization. For direct grafting, pieces tissue embryos (embryonic day 19, E21) newborn rats (PO) placed anterior eye chamber adult immediately dissection. grafting deep-freeze storage, taken (E16-E21) (PO), frozen a cooling rate 1 C/min CO2 N2 vapours addition cryoprotective agent dimethylsolfoxide (DMSO), stored for 33 days before thawing intraocular grafting. From 20 68 recipient sacrificed, their eyes sectioned, sections stained thionine cell bodies, Timm's sulphide silver method fiber systems terminal fields, acetylcholinesterase (AChE) cholinergic fibers AChE-positive neurons. When examining 101 (34 fresh 67 freeze-stored tissue) significantly lower survival was found (28 vs. 88%). The age-dependent no ages E16 PO, while E18-E21 had 50% rate. also smaller showed an increased tendency fragmentation. evaluating structure grafts, primarily have been harmful dentate granule cells precursors. organization intrinsic connections followed pattern known lesion intracerebral transplant studies. While demonstrating that brain can survive subsequent study indicates different schemes may be used get optimal neuronal populations.
