摘要: Objective To assess the effect of long-term pharmacological inhibition miR-21 in a model metabolic syndrome and obesity. Methods Aged db/db mice were treated with locked nucleic acid-modified anti-miRs directed against (LNA-21), control LNAs or PBS for 18 weeks. Cardiac function was assessed by echocardiography on body weight white adipose tissue (WAT) evaluated. Results MiR-21 expression efficiently inhibited heart WAT no apparent liver toxicity deterioration kidney function. MiR-21 had cardiac hypertrophy as well systolic diastolic functions. However, levels collagen 1 modestly reduced LNA-21 mice. weight, adipocyte size serum triglycerides significantly decreased. The targets TGFβ-receptor 2 (TGFBR2) phosphatase tensin homolog (PTEN) derepressed Sprouty1 increased after inhibition. Conclusions Long-term treatment is safe suppresses expression. not affected. led to significant loss reduces derepression TGFRB2, PTEN, 2.
