摘要: Determination of hepatitis D virus (HDV) genotypes is epidemiologically and clinically important. Phylogenic analysis based on sequencing multiple HDV strains isolated from sera patients not convenient for mass screening in routine laboratories. This study was designed to develop genotype-specific antibodies against delta antigen (HDAg) apply these immunohistochemical differentiation formalin-fixed, paraffin-embedded liver biopsies patients. Divergence the carboxyl-terminal 19 amino acids large HDAg between I II more than 70%. Peptides covering residues were conjugated keyhole limpet hemocyanin used immunization. The generated confirmed their specificity by binding type-specific HDAgs expressed DNA-transfected Huh-7 hepatoma cells. Liver 6 who had dominant genotype 33 stained with antibodies. accuracy 94.9%, agreement serum hepatic staining nearly perfect (κ = 0.83). In summary, can be as immunogens generate These proven useful biopsies.
