摘要: Abstract The aim of this study was to compare four HIV-1 viral quantitation platforms, Nuclisens EasyQ v2.0 ® (EQ), COBAS AmpliPreP/Cobas Taqman test v 2.0 (CTM), GENERIC HIV CHARGE VIRALE (GEN), with Abbott Real Time (m2000sp/rt) as reference technique. had first evaluated m2000sp/rt performances and then compared between techniques. Discordant samples were genotyped on gag pol gene sequences analyzed using Sequence locator SeqPublish detect eventual mismatches. Performance analysis showed good results coefficients variation values (CV) 1.35%, 0.65%, 0.54% for repeatability testing low, intermediate high concentrations, respectively. Reproducibility tests low CV 2.36% 1.42% concentration levels, respectively contamination very value 0.94%. Correlation concordance techniques ranged from r 2 = 0.98 bias = −0.00185 (for m 2000sp/rt vs CTM) = 0.90 bias = −0.135 EQ GEN). Discrepancies observed 37 mostly CRF02_AG but despite some mismatches, sequence (26/37) did not show any remarkable differences queries references. This correlation However, yielded under-quantitation CRF02_AG.
