摘要: Publisher Summary This chapter focuses on the various methods used to label cells with arachidonic acid and quantify arachidonate release from phospholipids in human neutrophil. Various assumptions problems are encountered when using these emphasized. These include effects of concentration incubation time ability obtain equilibrium labeling conditions, loss labeled phospholipids, formation radiolabeled eicosanoids. The major problem is nonuniform uptake exogenous into phospholipid molecular species. In neutrophil, its distribution complex lipids greatly influenced by (1) amount provided cell, (2) exposure acid. experiments presented demonstrates that even after 120 min incubation, there can be differences as great 10-fold between radiospecific activity Another assumption made performing or mass studies a equated being source for chapter, leukotrienes produced during cell activation represent only small fraction total lost all phospholipids.
